Studies on the Tuberculin

Abstract
Koch¢¥ s Old Tuberculin (O. T.) has been widely used in Korea for the eradication of animal tuberculosis since 1913. This O. T. was produced by use of glycerol beef broth. Therefore, the unused nitrogenous protein contained in this tuberculin was frequently blamed for the undesirable reaction in animal.
To eliminate this undesirable substances, Sauton¢¥ s synthetic medium was adopted and produced an experimental Heat Concentrated Synthetic Medium Tuberculin (HCSM Tuberculin). In addition, the comparisons were made between glycerol beef broth and Sauton¢¥ s synthetic medium of the growth pattern and pellicle formation of the various strains of Mycobacterium tuberculosis as well as tuberculin reactivity on guinea pigs sensitized with killed organisms of Pn, Dt, and C strains. The results are summarized as follows:
1) Pellicle formation of the organisms on the Sauton¢¥ s potato medium was much favorable rather than that on the glycerol beef broth, and 14 days after seeding the organisms, thin pellicle could be transferable to glycerol beef broth medium and Sauton¢¥ s synthetic medium.
2) Twelve lots of tuberculin (5 lots of glycerol beef broth origin and 7 lots of Sauton¢¥ s medium origin) were produced and tested their reactivity on the sensitized guinea pigs.
Tuberculin reactivity of 5 lots of glycerol beef broth origin was under the threshold- of=$5%, while of 4 lots of Sauton¢¥s synthetic medium origin, three were in 85 to 125% of confidence limit, and the other was more than 125%.
3) Statistical analysis on the tuberculin reactivity was made with 3 lots of Sauton¢¥ s medium origin of which confidence limit was within 85 to 125%, but the significant difference was not observed in the level of 5%.
4) There was no difference of the reading on 24 and 48 hours after intradermic injection of the O. T and HCSM Tuberculin. But local reaction was much lasted in injection site of HCSM Tuberculin.
